Succinate accumulation in vivo following injection of malonate.

چکیده
Among the factors involved in the accumulation of citrate by a tissue following the injection of fluoroacetate in viva is the ability of the tissue to convert precursors into 4-carbon acids (1). An indirect measure of the importance of this conversion was reported in a recent study (2) in which the Krebs cycle was blocked both by fluoroacetate and malonate, but in this study concurrent analyses for succinate were not attempted. Recently developed chromatographic techniques (3) have made it possible to determine not only the concentration of succinate, the metabolite accumulated, but also the concentration of malonate, the inhibitor, in tissue extracts following the injection of malonate. Previous studies on the effects of malonate in wivo have dealt with the metabolism of CY-malonate (4-6) and the effects of malonate on components of blood and urine (7-10). In the present study, the optimal conditions for the accumulation of succinate in tissues in viva were defined, and the rate of excretion of succinate, citrate, and malonate in the urine was determined.

Studies on the conversion of acetate, lactate, and malonate to succinate in the intact rat.
This investigation, preliminary reports on parts of which have previously appeared (1, 2), presents data on the conversion in the intact rat of the labeled carbon of the following compounds to succinate: NaHC*03, CH&*OOH, C*H&*OOH, CH&HOHC*OOH, and C*H&*HOHCOOH. All the animals were injected with malonate to augment the urinary excretion of succinate (3). The latter was isolated from the urine ...
متن کاملSIGNIFICANT CHANGES IN THE ACTIVITY OF GABATRANSAMINASE AND SUCCINATE SEMIALDEHYDE DEHYDROGENASE OF MOUSE HYPOTHALAMUS FOLLOWING PERIPHERAL INJECTION OF CHOLECYSTOKININ-8 AND/OR CAERULEIN
The activities of 4-aminobutyric-2-oxoglutaric acid transaminase (GABA-T) and succinate semialdehyde dehydrogenase (SSADH) were determined in mouse hypothalamus after peripheral injections of cholecystokinin-8 (CCK-X)and/or caerulein (CLN). GABA transaminase activity was measured utilizing endogenous succinate semialdellyde dehydrogenase to convert the product of GAB A-T, succinate semiald...
متن کاملMalonate inhibition of oxidations in the Krebs tricarboxylic acid cycle.
The inhibition of succinate oxidation by malonate is a well known phenomenon. Since the oxidation of succinate to fumarate is an integral part of the Krebs cycle of oxidations, it has been generally assumed that the inhibitory effect of malonate upon the oxidation of any member of the cycle is the result of the inhibition of the succinate to fumarate step. However, the present paper provides ev...
متن کاملTransport of dicarboxylic acids in castor bean mitochondria.
Mitochondria from castor bean (Ricinus communis cv Hale) endosperm, purified on sucrose gradients, were used to investigate transport of dicarboxylic acids. The isolated mitochondria oxidized malate and succinate with respiratory control ratios greater than 2 and ADP/O ratios of 2.6 and 1.7, respectively. Net accumulation of (14)C from [(14)C]malate or [(14)C]succinate into the mitochondrial ma...
متن کاملCentral administration of methamphetamine synergizes with metabolic inhibition to deplete striatal monoamines.
These studies examined, in vivo, the effect of local intrastriatal perfusion of methamphetamine (MA) on dopamine (DA) and glutamate release in relation to changes in striatal DA and serotonin (5-HT) content measured 1 week after treatment. Interactions between the inhibition of energy metabolism and the direct perfusion of MA on long-term decreases in DA and 5-HT content also were investigated....
